In this study we describe a rapid procedure suitable for the identification of induced recessive and dominant mutations applied to two Zea mays mutants expressing a dwarf and a pale green phenotype, respectively, which were obtained through pollen EMS mutagenesis.DNA was extracted using the Qiagen DNeasy Plant mini Kit according to the manufacturer's protocol. In total 32 DNA samples were used for Illumina Sequencing. Thirty two Illumina uniquely barcoded paired-end (2 x 100 bp) libraries were generated from sheared DNA (fragment size 400 – 500 bp) as described by (Meyer and Kircher, 2010).
