A suppressor of the short-hypocotyl phenotype of shv3svl1 was mapped to suc1, encoding a plasma membrane Suc/H+ symporter. The EMS-induced suppressor mutant was backcrossed to the shv3svl1 progenitor and ~150 segregants were pooled from the BCF2 generation. DNA was extracted and SNP frequencies were determined in order to map the mutation. A second library was prepared with DNA from the shv3svl1 parent in order to identify and subtract SNPs present in shv3svl1 relative to the Col-0 reference sequence.
