To understand of the impact of KLF5 and NFYA on metabolism in prostate cancer cells LNCaP cells were plated in RPMI 1640 media with no phenol red and with 5% charcoal stripped serum, sodium pyruvate, penicillin and streptomycin. After 48h (to allow adnrogen deprivation), fresh media was added, with 96% ethanol or the synthetic androgen R1881 (10nM concentration). After trypsinization cells were seeded in media with CSS and transfected with a pool of non-targeted control siRNAs or a pool of siRNAs targeted against KLF5 and NFYA
