Sequencing of CRISPR edited EGFR mutant lung cancer cell lines

CRISPR-Cas9 was performed to introduce an in cis C797S mutation in H3255GR and DFCI52 human lung cancer cells which harbor the EGFR L858R/T790M mutation in order to assess the potency of JBJ-09-063 in the presence of the triple mutation. Changes in variant allele frequencies in bulk cells treated with osimertinib were analyzed by CRISPR sequencing and JBJ-09-063 potency was evaluated in vitro and in vivo as a single agent and in combination treatment using these models.
