Evaluation of transcriptional changes of bone marrow derived macrophages (BMDM) in response to necrotic pancreatic acinar cells

To investigate the activation and polarisation of BMDMs during the course of pancreatitis, BMDMs were co-incubated with CCK-stimulated acinar cells (+/- the serine protease inhibitor nafamostat) for 6h in an in vitro experiment. The LPS treatment served as a positive control. We used BMDMs isolated from C57Bl/6 mice which were maintained in the presence of 20ng MCSF over 7d until they were confluent. Co-incubation was performed with freshly prepared acinar cells from C57Bl/6 mice over 6h +/- Nafamostat. BMDMs were carefully washed before RNA Isolation. Transcriptome analyses were performed using individual RNA samples.
