The goal was to assess patterns of gene flow and genetic structuring of Fundulus olivaceus within study watersheds, related to environmental factors which might affect gene flow. The specific focus was on testing how watershed geomorphic evolution drives changes to habitat and dispersal corridor quality, affecting metapopulation structure. Fundulus olivaceus was chosen for a study organism as it relies on habitats created by fluvial geomorphic processes (pools and backwaters), but is not directly tied to sediment caliber. Five to ten individuals were collected from 3-7 headwater sites per watershed (n = 17 watersheds, total fishes = 721). Sample libraries were prepped with the Eco-T221 restriction enzyme and sequenced on an Illumina HiSeq platform.
