miRNA and mRNA transcriptome profiling of chondrocytes in proliferative zone of acetabular cartilage in developmental dysplasia of the hip

Developmental dysplasia of the hip (DDH) is a complex and multifactorial disease with an unclear pathogenesis, and the non-coding RNAs have been found important effect in many diseases. However, their contributions to the pathogenesis of DDH are not well-established. Growth of the acetabular cartilage mainly depends on the regulation of chondrocytes in proliferative zone. Here, we report on some new regulatory mechanisms of gene expression underlying the development of DDH by transcriptome profiling of proliferative zone microdissected from the acetabular cartilage. These results provide original insight into the roles of non-coding RNAs in DDH and provide a valuable resource for further analyses of molecular mechanisms and signaling networks. Overall design: We explored the mechanism underlying swadding-induced DDH by profiling mRNA and miRNAs expression via RNA-seq. Six neonatal rats were swaddled with use of surgical tape after birth in a manner simulating the human practice as DDH group, and other six neonatal rats in same litter were as normal control. The acetabular cartilage were obtained on five and ten days after birth. Total RNA from microdissected material of the acetabular cartilage was extracted according to the instruction manual of the TRlzol Reagent (Life technologies, California, USA) and the RNA integrity was assessed using the RNA Nano 6000 Assay Kit of the Agilent Bioanalyzer 2100 System (Agilent Technologies, CA, USA). A total amount of 1.5 µg RNA per sample was used as input material for Library preparation and sequencing.
