Phase Ib/II Study of Azacitidine, Venetoclax and Magrolimab in Adult Patients with Newly Diagnosed and Relapsed Refractory Acute Myeloid Leukemia

To better understand the mechanisms of response and resistance to azacitdine, venetoclax, and magrolimab (AVM), we performed single-cell RNA-sequencing (scRNAseq) on 27 samples (11 pre- and 16 post-treatment) from 11 TP53-mutated patients with combined therapy Overall design: Bone marrow biopsies were routinely collected. Samples were freshly frozen with freezing medium containing 20% fetal calf serum (FCS) and 10% dimethyl sulfoxide (DMSO) in Dulbecco's modified Eagle medium (DMEM) and stored in liquid nitrogen. All frozen BM samples were retrieved immediately before sample processing. The 5 prime gene expression libraries were prepared using a 10x Genomics Chromium Controller instrument and Chromium Single-Cell 5 V5.1 reagent kits (10x Genomics).
