To identify differentially expressed gene in the atrium tissues between wildtype control and the Dnajb6 heterozygous knockout mouse

To seek molecular mechanisms underlying the sick sinu syndrome phenotypes observed in Dnajb6 heterozygous knockout mice, we performed whole transcriptome RNA-sequencing experiments using right atrial tissues isolated from Dnajb6 heterozygous knockout mic and WT control mice at 1 year of age. Total RNA was extracted from the right atrium (RA) tissues of 1-year-old Dnajb6+/- heterozygous mutant hearts and WT sibling controls. Six total samples (3 biological replicates for each genotype) were submitted for RNA sequencing (Azenta Life Science, NJ). Genes were considered to be differentially expressed between the two groups if they exhibited a greater than 2-fold change and an False Discovery Rate (FDR) of less than 0.05 according to the DESeq approach.

为探究Dnajb6杂合敲除小鼠中观察到的病窦综合征(sick sinus syndrome)表型的潜在分子机制,我们以1岁龄Dnajb6杂合敲除小鼠及野生型(Wild Type, WT)对照小鼠的分离右心房组织为材料开展了全转录组RNA测序实验。我们从1岁龄Dnajb6+/-杂合突变小鼠的心脏右心房(right atrium, RA)组织,以及同窝野生型对照小鼠的对应组织中提取了总RNA。最终共制备6份样本(两种基因型各3份生物学重复),并将其送至美国新泽西州的Azenta生命科学公司(Azenta Life Science, NJ)进行RNA测序。采用DESeq方法进行差异表达分析后,若基因的表达变化倍数大于2倍且错误发现率(False Discovery Rate, FDR)小于0.05,则判定该基因在两组间存在差异表达。
