Analysis of planktonic fungi during spring phytoplankton bloom at Helgoland Roads (German Bight, North Sea) in 2017

41 surface water samples were collected from March to May 2017 during the course of a spring phytoplankton bloom at the LTER Helgoland Roads, German Bight (North Sea). Samples were sequentially filtered through 10, 3 and 0.2 micrometer polycarbonate filters. The DNA was extracted from the 0.2 and 3 micrometer fraction using a chloroform:phenol approach. The extracted DNA was amplified using the primers published in Herlemann, 2011 (Baltic Sea study). The primers were Illumina primers and contained the standard MiSeq adaptors and barcodes. The amplified products were cleaned using ampure beads and quality checked on a fragment analyser. 0.2 and 3 micrometer amplified products from the same sample were pooled together in equimolar concentration to represent the entire community from 0.2 - 10 micrometer. All samples were then pooled into libraries and sequenced using an Illumina MiSeq 2500 (2x 250 bp)
