Transcriptome comparison of PMN-MDSCs and and normal neutrophils

Polymorphonuclear myeloid-derived suppressor cell (PMN-MDSC), also named pathologically activated neutrophil, is a critical component of tumor microenvironment (TME), playing crucial roles in tumor progression and therapy resistance. Here, we compared the transcriptome of PMN-MDSCs from B16 tumor bearing mice and the neutrophils from tumor free mice. B16-F10 melanoma cells were inoculated subcutaneously into wild-type C57/BL6 mice. At day 15 post inoculation, PMN-MDSCs were isolated spleen of tumor bearing (TB) mice by AutoMACS with EasySep Mouse Neutrophil Enrichment Kit (Stem cell). Normal neutrophils were isolated from tumor free (TF) heathy control mice by the same methods. Total RNA was extracted by TRIZOL (Invitrogen) for RNAseq and comparison.
