A salt-sensitive cucumber (Cucumis sativus L.) cv. Jinchun No. 2, and a salt-tolerant pumpkin (C. maxima × C. moschata) cv. Chaojiquanwang was used in this experiment .Seeds were surface sterilized with 1% HClO for 30 min and rinsed thoroughly with distilled water, and then incubated in the dark at 26°C until germination. Germinated seeds were then directly grown hydroponically in aerated basic salt media (BSM) solution (0.5 mM KCl, 0.1 mM CaCl2, pH 5.7 non-buffered) in the dark for 5-6 days at room temperature (24°C),Then the seedlings were subjected to 75 mM NaCl stress for a period of 24 hours,using 0 mM NaCl as control,Fresh root apexes (0-2 mm from the root tip) were harvested for RNA_seq experiment,3 biological repelicates for each treatment .
