Single cell RNA sequencing of ischemia reperfused kidneys from Cre- and endothelial cell specific prolylhydroxylase 1, 2, and 3 knockout mice

we performed scRNA sequening on day14 post renal IRI kidneys from a mutant mouse with acute inactivation of endothelial PHD1, PHD2 and PHD3 compared to control Overall design: 8-10 week old Cre- and Cdh5-CreERT2;Phd1f/f, Phd2f/f, Phd3f/f mice were subjected to 25 minute ischemia reperfusion injury (IRI) at 37°C. Cre-mediated inactivation was induced by four intraperitoneal (IP) injections of tamoxifen dissolved in 10% ethanol in corn oil at 20 mg/mL (3 mg/mouse) given every other day.On post IRI day 14, IR kidneys were isolated and processed for scRNA seq analysis.
