In order to search for cell-intrinsic targets for Brg1 in ILC3s, Brg1-deficient and control ILC3s were sorted from bone marrow chimeric mice, generated by transfering mixed bone marrow cells of Thy1.1/Thy1.2Rag1-/-Smarca4f/f and Thy1.2Rag1-/-Smarca4f/fRorc-cre donors at 1:1 ratio to sub-lethally irriadiated Rag2-/-Il2rg-/- recipients. Overall design: 2.5 million bone marrow cells from Thy1.2Rag1-/-Smarca4f/f and Thy1.1Thy1.2Rag1-/-Smarca4f/fRorc-cre mice respectively were mixed and transferred to sub-lethally irradiated Rag2-/-Il2rg-/- mice. Small intestinal lamina propria lymphocytes were isolated from the chimeric Rag2-/-Il2rg-/- mice 6-7 weeks later. ILC3s from the Thy1.2Rag1-/-Smarca4f/f origin (Thy1.2highThy1.1-CD45low) and the Thy1.1Thy1.2Rag1-/-Smarca4f/fRorc-cre origin (Thy1.2highThy1.1+CD45low) were sorted by flow cytometry. mRNA was extracted by Trizol reagent and RNA-seq analyses were performed.
