Failure to precisely repair DNA damage in self-renewing Hematopoietic Stem and early Progenitor Cells (HSPCs) can disrupt normal hematopoiesis and promote leukemogenesis. HSPCs are widely considered a target of ionizing radiation (IR)-induced hematopoietic injury. In this project, we are studying conditions that can protect the HSPCs from IR. Overall design: The experiment was done in three types of media and in two time-points (4h and 24h). Type of medium is: plain medium, medium with Mouse bone marrow fluid, and medium with Mouse bone marrow cells. each type was treated with IR for 4h or 24h and each has a control. Total samples: 3 medium types X (4 biological replicates X 2-time points X 2 (control and treated)) = 48 samples.
