Ghrelin deletion and conditional ghrelin-cell ablation increase pancreatic islet size in mice

We utilized single cell sequencing of FACS sorted cells from pancreatic islets of wildtype and ghrelin knock out mice to understand the effects of ghrelin deletion on gene expression profiles of various islet cells. Overall design: Single-cell RNA sequencing (10X Genomics) was performed on FACS sorted viable cells from the pancreatic islets of wildtype and ghrelin knockout (homogyous) mice. For each genotype, pancreas from 4 mice were pooled for single-cell dissociation, followed by FACS sorting and 10X single-cell RNA sequencing.
