STARR-seq assay tiling across putative hominoid-specific enhancers and for orthologs from 11 primates and 9 ancestral reconstructions of each active tile Three 60mm plates, each with about 1.5 million HepG2 cells, were transfected with 5ug of either our tiling library or ortholog library and 2.5ug of a puromycin expressing palsmid. Cells were selected with 1ng/mL puromycin for 24 hours and then harvested for RNA and DNA extraciton.
