De novo generation of macrophage from placenta-derived hemogenic endothelium

We profile the transcriptional landscape of mouse fetal placenta and identify four distinct types of haematopoietic stem/progenitor cells (HSPCs) during mid-gestation using single-cell RNA sequencing. We experimentally validate and uncover that a subpopulation of placental endothelial cells exhibits hemogenic endothelial (HE) potential and shares transcriptional features with HE cells in the aorta-gonad-mesonephros (AGM) region. Examination of gene expressive levels in sorting placental cells, placental HE cells, AGM HE cells, placental ECs, AGM ECs and yolk sac ECs from pregnant mice.
本研究解析了小鼠胎儿胎盘的转录组图谱,并借助单细胞RNA测序技术,鉴定出妊娠中期存在的四种不同造血干/祖细胞(haematopoietic stem/progenitor cells, HSPCs)亚型。经实验验证并揭示,胎盘内皮细胞的一个亚群具有生血内皮细胞(hemogenic endothelial, HE)潜能,且与主动脉-性腺-中肾(aorta-gonad-mesonephros, AGM)区域的生血内皮细胞共享转录组特征。本研究还对妊娠小鼠来源的分选胎盘细胞、胎盘生血内皮细胞、AGM生血内皮细胞、胎盘内皮细胞(endothelial cells, ECs)、AGM内皮细胞以及卵黄囊内皮细胞的基因表达水平进行了检测分析。
