The RNA-seq data was generated to evaluate the efforts of elevated mTOR signaling on the inflammatory transcriptome in the mouse microglia. Our data shows that there were no significant differences in expression levels of major proinflammatory cytokines, chemokines and inflammatory mediators. However, there was a significant induction of lysosome genes, along with up- or down-regulation of genes involved in phagocytosis in TSC1 knockout microglia. Mouse brains were harvested from four pairs of wildtype and TSC1Cx3cr1CKO mice at 4 weeks of age. Microglia were freshly isolated utilizing anti-Cx3cr1-PE antibody (Biolegend), coupled with anti-PE MicroBeads (Miltenyi). Microglia were finally captured using magnetic MS column (Miltenyi). Approximately 96.8 % of purified cells were CD11b positive. Total RNA samples were immediately prepared from purified cells using Trizol method.
