Short-term culture with IL-7 improves CRISPR/Cas9-mediated gene-editing efficiency in naïve T cells. In order to investigate the effect of culture with IL-7, we performed RNA sequencing analysis using human T cells. Overall design: Human peripheral blood mononuclear cells derived from 2 healthy different donors (0309 and D110) were used. CD8+ naïve T cells (CD8+CD45+CCR7+), CD8+ unstimulated T cells cultured in the presence of IL-7 for 3 days (CD8+CD45+CCR7+), and CD8+ T cells which were stimulated and cultured in the presence of IL-2 and IL-15 for 3 days (CD8+) were isolated by flow cytometry and subjected to RNA sequencing.
