PIWI-interacting RNA expression regulates pathogenesis in a Caenorhabditis elegans model of Lewy body disease

PIWI-interacting RNAs (piRNAs) are small noncoding RNAs that regulate gene expression, yet their molecular functions in neurobiology are unclear. Our results find that dysregulated piRNAs upregulated H3K9me3 to silence gene expression in human Alpha-syn(A53T) and A_beta;Alpha-syn(A53T) pan-neuronal overexpressing strains. To explore which gene was modified, we carried out Chip-Seq in C. elegans including wild type (N2), A_beta strain (CL2355), Alpha-syn(A53T) strain (UM0010), A_beta;Alpha-syn(A53T) strain (UM0022), tofu-1 mutant (tm6424), Alpha-syn(A53T);tofu-1 strain (UM0028), A_beta;Alpha-syn(A53T);tofu-1 strain (UM0029) to get the genes modified by H3K9me3. Our samples were sequenced in two lanes by single end, which should combine two lane data in analysis.
