DPM1 regulates ER-stress sensor IRE1 activity and antitumor immunity in colorectal cancer

This project investigates how DPM1 regulates the ER stress sensor IRE1 and how this impacts tumor immunogenicity and response to immune checkpoint blockade in colorectal cancer models. We profile tumor immune microenvironments and T cell receptor repertoires in wild-type and Dpm1-deficient mouse tumors to define IRE1-dependent immune programs and mechanisms of enhanced antitumor immunity. Single cell data were generated using the 10x Genomics Chromium platform with 5' gene expression, feature barcoding for hashtags, and paired TCR VDJ libraries. Data were processed using Cell Ranger multi (v7.2.0) and downstream clonotype and joint analyses were performed with CONGA.
