This study aims to expand our knowledge of the genome-wide positioning of yeast nucleosomes. Isogenic yeast strains expressing H2A-A122C, H4-S47C or neither were used to conduct chemical mapping and MNase-seq. Isolated nuclei were used for the chemical cleavage and MNase digestion. Both cysteine mutations caused cleavage of DNA at specific sites in the nucleosome structure, allowing us to identify nucleosome positions at base-pair resolution.
