Described experiments are published in https://doi.org/10.1002/advs.202307695. Two murine PDAC cell lines were transduced with pLenti-PGK-puro-Prkaa1 and empty control plasmids to generate Prkaa1 overexpressing cell lines (8182_PGK_empty vs. 8182_PGK_Prkaa1, 9091_PGK_empty vs. 9091_PGK_Prkaa1, Addgene #19068 and #204356). Furthermore, three murine PDAC cell lines were used to generate Prkaa1 KO cell lines. For each cell line, one control plasmid (LacZ) and two different sgRNAs targeting Prkaa1 were used (8570_LacZ vs. 8570_Prkaa1_KO1 vs. 8570_Prkaa1_KO2, 9091_LacZ vs. 9091_Prkaa1_KO1 vs. 9091_Prkaa1_KO2, 8248_LacZ vs. 8248_Prkaa1_KO1 vs. 8248_Prkaa1_KO2, doi: 10.1038/s43018-021-00326-1). Finally, two murine PDAC cell lines were treated with PF-3758309 for 24 hours (8182_con vs. 8182_PF3758309, 9091_con vs. 9091_PF3758309).
