Firstly, we screened donepezil as a positive compound that can promote NPC-derived OPCs to Ols differentiation from a small library containing many old drugs and pathway regulators using this differentiation assay. Sencondly, we furthure found that it can promote primary OPCs to differentiate into OLs and enhance meylination in OPC-DRG neuron co-culture systerm in vito. Finally, we examined the efficacy of donepezil in enhancing remyelination in vivo in the cuprizoneinduced demyelination model. Samples included negative control, positive control, and donepezil treatment group. Immunofluorescence staining, Q-PCR, immunohistochemistry and scanning electron microscopy were used to obtain reliable data.
