Hepatocyte-Specific Transcriptional Profiling of the Hepatic Fasting Response

We generated transcriptional profiles of fasting-induced changes of in vivo hepatocytes using the INTACT methodology. We subjected female HEP-INTACT mice to an overnight fast starting at the onset of the mouse active phase, ZT12, and after 16h livers were collected for INTACT followed by RNA-seq analyses of isolated hepatocytes. Ad libitum fed control mice were sacrificed at the same time point to account for diurnal variations in gene expression.
本研究采用INTACT技术,构建了活体肝细胞禁食诱导变化的转录组图谱。我们将雌性HEP-INTACT小鼠于其活动周期起始点ZT12启动过夜禁食,禁食16小时后采集肝脏样本,经INTACT技术分离肝细胞后,对分离得到的肝细胞开展RNA-seq分析。为校正基因表达的昼夜节律波动,我们于相同时间点处死自由采食的对照组小鼠。
