Due to a lack of available MRG15 antibody for ChIP-seq analysis, exogenous liver expression of a FLAG-MRG15 under a liver-specific TBG (thyroxine-binding globulin) promoter was executed using AAV vectors, and ChIP-seq was conducted using FLAG antibody. To examine possible dynamic changes in binding activity of MRG15, liver tissues were collected for analysis at both ZT10 [where ZT is Zeitgeber time (light on at ZT0 and off at ZT12)], the light period when mice are inactive and less fed; and ZT22, the dark period when mice are active and feeding. Liver tissues infected with AAV-luciferase viruses were used as control.
