Pathogenic Stem Cell Variation in Chronic Lung Disease

Chronic obstructive lung disease is the third leading cause of death in the United States. We have identified and cloned the stem cell variants from patients lung samples using our cloning method. These stem cells functions are explored using combinations of microarray, single cell RNA-seq, ChIP-seq and in vivo assays. We have cultured distal lung stem cells colony on 3T3 feeder layer. The stem cells are injected into mouse for the in vivo differentiation and mouse xenograft is used to examine the functional aspect of the stem cells. Microarray is performed for the stem cells and cells from the xenograft (Xeno) samples using Affymetrix human exon 1.0 st array chip.
