Allergic rhinitis is a prevalent condition affecting up to 25% of the global population. The role of innate lymphoid cells in the pathogenesis of allergic diseases have been previously described, however, a single cell transcriptomic analysis to investigate differences between allergic and non-atopic healthy individuals have yet to be fully investigated. Using a combination of single cell RNA/protein and FACS validation, we provide evidence that there is a predominance and dysregulation of a type 2 response in the ILCs of allergic patients. Overall design: Single cell RNA sequencing was performed by isolating human innate lymphoid cells (ILCs) by FACS sorting from peripheral blood mononuclear cells. Libraries (3' GEX and protein libraries) were prepared using Chromium single cell 3' Reagent Kits according to manufacturer's protocol (10x Genomics). The generated scRNAseq libraries were sequenced using HiSeq 4000 sequencers. Human samples were processed and sequenced for allergic group (n=2) and non-allergic healthy control (n=1).
