Multiple epiphyseal dysplasia (MED) results from mutations in cartilage structural proteins COMP, MATN3 and type IX collage. V194D Matn3 is a mouse model of MED in which a signifficant contribution of ER stress has been detected. In order to test whether ER stress contribution was a beneficial or detrimental component of the disease, V194D Matn3 mice were crossed with a cartilage specific knock-out of Xbp1, and transcriptomic analysis was performed using the Affymetrix 420A Mouse Array. Overall design: Chondrocytes were extracted from postnatal day 5 ribs of Xbp1 WT and KO mice on V194D Matn3 or WT Matn3 background. RNA was extracted from isolated chondrocytes using Trizol reagentand pooled from 3 separate extractions. A GeneChip 3’ expression assay (Mouse430_2 Affymetrix) was used to analyse gene expression. Quality control checks for control hybridizations were performed using Microarray Suite 5.
