This dataset features transcriptomic profiles of feeder-free extended pluripotent stem cells (ffEPSCs) and their parentalderived from human embryonic stem cells (ESCs). ffEPSCs were generated using a Matrigel-based feeder-free system combined with a chemical cocktail to convert conventional ESCs into ffEPSCs. Generated using Smart-seq2 and sequenced on Illumina HiSeq 2000, the dataset explores transcriptional profiles and heterogeneity of ESCs and ffEPSCs, providing insights into early human development and stem cell biology. The samples in this submission include human embryonic stem cells (ESCs) and extended pluripotent stem cells (ffEPSCs) derived from ESCs. Specifically, the human ESC line H9 was used for conversion into ffEPSCs. Both ESCs and ffEPSCs were analyzed at the single-cell level using Smart-seq2 sequencing. Replicates were included, as the experiments were performed on multiple cells from each condition. There are reference samples (ESCs) as well as experimental samples (ffEPSCs) to investigate the transition from ESC to ffEPSC. The primary variables under investigation are the differences in gene expression between these two states under controlled experimental conditions.
