The emergence of cellular resistances to oncolytic viruses is a largely unexplored process. Here, we isolated and characterized mouse melanoma cells (B16-F10) that became resistant to an oncolytic vesicular stomatitis virus (VSV-D51). RNA-seq analysis demonstrated that resistance was associated to extensive changes in gene expression patterns, with hundreds of differentially expressed genes. The observed gene expression shifts were highly correlated between different VSV-resistant subclones, suggesting that resistance to oncolytic virus treatment is a conserved and predictable process.
