The role of POLR2A/RPB1 in kidney proximal tubule responses to DNA damage-induced acute kidney injury is unknown. We undertook treatment of HK2 cells with the DNA damaging agent aristolochic acid, with or without knockdown of POLR2A to determine the role of RPB1 in kidney cells. Overall design: HK-2 cells were transfected with 10nM Trilencer-27 siRNA duplexes specific to POLR2A or Trilencer-27 universal scrambled control siRNA duplex (Origene), using SiTran2.0 transfection reagent (Origene), according to manufacturer's instructions. At the same time, cells were also treated with DMSO (control) or 25µM AAI for 24 hours.
