Single-cell RNA sequencing of PBMCs from patients with myasthenia gravis and healthy controls

T and B cells from PBMCs were labeled and processed for single-cell capture using the BD Rhapsody system. Overall design: Peripheral blood mononuclear cells (PBMCs) were isolated from patients with myasthenia gravis and healthy controls. T cells and B cells were enriched and subjected to scRNA-seq. Simultaneously, surface protein expression was profiled using Abseq. In a subset of samples, immune repertoire analysis was also performed. Several samples were multiplexed using antibody-derived tags, and demultiplexing was conducted prior to downstream analyses.
