These experiments aim to detect changes in the transcript (mRNA) of cardiac tissue when animals are exposed to high temperatures and heat stroke occurs due to the increase in environmental temperature. The research objectives include: 1) detecting changes in transcripts (mRNA) of heart tissue during heat stroke caused by elevated environmental temperatures; 2) Detecting transcripts that can serve as biomarkers for heat stroke induced myocardial injury. This study used 6–8 weeks C57BL/6N male mice (body weight: 20–25 g). All mice were acclimated for 1 week under standard housing conditions: 22–25°C with a 12-hour light/dark cycle, and free access to food and water.The sham group was maintained in a normal environment (22–25°C, 60–65% humidity) with fasting and water deprivation. The HS group was exposed to high temperature intervention in a climate controlled chamber, with the temperature set to 40±0.5°C and humidity maintained at 60–65%. During this period, the mice were fasted and water-deprived. Rectal temperature was measured every 30 minutes using a rectal thermometer until the core body temperature reached 42°C, at which point the mice were immediately transferred to a recovery environment (22–25°C, 60–65% humidity).After confirming the successful modeling, we collected mouse heart tissue for further experiments.
