Overexpression of high levels of human UHRF1 tagged with EGFP (hUHRF1-EGFP)high in zebrafish hepatocytes induces cellular senescence by 5 days post fertilization (dpf) and hepatocellular carcinoma by 15-20 dpf. The goal of this study is to compare the transcriptome profile of zebrafish livers with (hUHRF1-EGFP)high overexpression compared to WT siblings. RNAseq was performed on RNA extracted from pools of microdissected liveres by using RiboZero libary prep to capture all transcripts, including transposable elements, that are expressed in the livers. Tg(fabp10a:hUHRF1-EGFO)high+/- were incrossed and the resulting larvae were raised and the liver from 20-30 larvae was manually dissected from 5 independent clutches at 120 hpf. Livers with UHRF1 overexpression were selected based on EGFP fluorescence and non-transgenic siblings with no fluorescence were collected as controls. Livers were manually dissected under the stereomicroscope and put directly in Trizol for total RNA extraction. RNA was DNAseI treated, depleted for ribosomal RNA by using Ribo-Zero Gold treatment and used for Illumina Truseq RNA sample prep kit, version 2.
