The mechanistic basis for interprotomer deglycosylation of antibodies by Corynebacterial IgG specific endoglycosidases

We made barcoded libraries of IgG1 Fc, each containing ~4,350 distinct single amino acid mutations. We then applied deep mutational scanning technology to quantify IgG1 Fc mutational impact on specificity and affinity for various endoglycosidases. We used a lentiviral system for expression of IgG1(216-447) on the surface of mammalian HEK293T cells.
