A single-cell atlas of bobtail squid visual and nervous system highlights molecular principles of convergent evolution

RNA-seq and scRNA-seq of dissected organs from adult and hatchling-stage cephalopod Euprymna berryi Overall design: Animals were euthanised using 4% ethanol in natural seawater. Organs were dissected and Total RNA extraction was carried out using TRIzol reagent (Invitrogen) for lysis followed by on-column purification using PureLink RNA Mini Kit (Invitrogen) as described in TRIzol Plus RNA Purification instructions from manufacturer. Demultiplexing was carried out using bcl2fastq (v2.19). scRNA-seq was carried out using 10x Genomics protocols for v.2 and v.3.1 kits as per manufacturer's instructions.
