ISG induction differs immunogenical human splenic cDC2 activation from homeostatic ones

To assess the commonalities and differences in the transcriptomic reprogramming involved in immunogenic and homeostatic (DC2hm) human cDC2 activation, we conducted a comparison of gene regulation programs between DC2hm and immunogenically activated cDC2s (Generated using TLR-agonist cocktails or LPS+IFN-?) by RNA-seq and found that though both programs upregulate common DC maturation markers like CCR7, CD80, CD40, and LAMP3, several transcription modules including ISG induction distinguish immunogenically activated cDC2s from DC2hm. Overall design: Human splenic cDC2s was stimulated in U-bottom 96-well plates for 12 h in complete medium with the combination of TLR-agonist including polyinosinic-polycytidylic acid [poly(I:C); 10 µg/ml], R848 (1 µg/ml), and LPS (4 µg/ml) or LPS (4 µg/mL) plus IFN-? (100 ng/mL).
