Immune Editing Roles of SIGLEC15 in Tumor Microenvironment [scRNA-seq]

SIGLEC15 in tumor microenvironment could dampen cytotoxic functions of T cells, and we blocked SIGLEC15 with mono-antibody to examine functional changes of cell populations in comparison to treatment of IgG. Overall design: Percutaneous tumor grafts from C57 mice, treated by IgG (n=3) or SIGLEC15 (n=3) antibody, were trimmed and digested for cell pelletes, and CD45 positive selection magnetic beads were used to enrich CD45+ cells. After examination of cell viability (>90%), CD45+ cells and unsorted cell pelletes were loaded for sample preparation at the ratio of 1:1.
