For functional studies, we worked with Enterococcus faecalis strain VE14089, derived from the sequenced strain V583, cured of its plasmids to improve genetic amenability. Although strain VE14089 has no major DNA rearrangements, with the exception of a ~20 kb integrated region of pTEF1 plasmid, the strain presented significant growth differences compared to V583. Genome sequencing was performed on VE14089. The aim of our study was to understand the molecular basis of the growth defect and to restore this defect for molecular functional studies of a representative member of the CC2 clonal complex.
