The overall goal of this project is to identify transcriptome alteration in fetal islets exposed to transient maternal hyperglycemia from GD20 to GD22. Left uterine horn fetus rats were subjected hyperglycemic infusion, while right uterine horn normoglycemic fetuses were used as paired internal controls. Upon completing glucose infusion from GD20 to 22, 2 fetal pancreata were pooled and islets were extracted via collagenase digestion and hand picked. After 24 hour of recovery in RPMI containing 5mM glucose, RNA was collected from the islets and sequenced using HiSeq4000.
