scRNAseq screening of meiosis-promoting factors

A barcoded PiggyBac transposon expression plasmid library of 88 candidate regulatory factors was screened for ability to induce meiosis in human induced pluripotent stem cells. scRNAseq was performed in order to characterize gene expression and identify meiotic cells. Barcode capture was performed by hybridizing the scRNAseq library with a biotinylated probe, resulting in a sublibrary enriched for barcode sequences. This sublibrary was also sequenced to identify which transgenes were expressed in each cell.
