The complete genome and methylome of Pseudomonas mendocina NEB698, the source of PmeI restriction endonuclease, was determined using PacBio sequencing and methylation analysis. Unlike other Type II restriction-modification systems, we found no methyltransferase partner for the PmeI endonuclease. The absence of host-protective methylation is made possible by an absence of any PmeI recognition sites (GTTTAAAC) within the genome.
