To investigate the effects of gE, gI and TK gene deletions on PRV, we constructed a gE/gI/TK triple gene deletion strain SD-2017gE/gI/TK using homologous recombination technology. Transcriptomic profiling was performed on PK-15 cells infected with SD-2017gE/gI/TK, parental SD-2017 strain, Bartha-K61 strain. The results showed that SD-2017gE/gI/TK had significantly downregulated expression of cytoskeletal genes involved in cell adhesion such as actin and connexin, as well as genes related to endoplasmic reticulum stress and protein degradation pathways like UPR response, compared to SD-2017 and Bartha-K61 strains. This study focused on observing the remarkable differences in virulence and invasion related genes between the deletion strain SD-2017gE/gI/TK and its parental and reference strains.
