N6 Methyladenosine is a reversible and common post transcriptional modification in mRNA. It regulates immune cell development and is involved in diseases like infections, autoimmune diseases, cardiovascular diseases, tumors, and liver diseases. However, the role of m6A in LPS induced macrophage inflammation is still unclear. In this study, we investigated the role of Zc3h13 in LPS induced liver inflammation. Macrophages, essential for the immune system, play a key role in liver injury. Our findings show that Zc3h13 is highly expressed in liver macrophages during LPS induced liver injury and is involved in inflammatory factor expression and macrophage polarization. In vivo studies indicate that knocking down Zc3h13 reduces LPS induced liver inflammation in mice. In vitro, silencing Zc3h13 in LPS induced macrophages decreases m6A methylation, reduces inflammation, and promotes macrophage polarization from M1 to M2. Additionally, Zc3h13 silencing stabilizes Spic mRNA in an m6A dependent manner, inhibiting NF kB pathway activation and reducing inflammation. In summary, our study suggests that silencing Zc3h13 reduces LPS induced liver injury by regulating the NF kB pathway in macrophages.
