To identify the biological functions of the PTEN R14M mutation, we performed transcriptome analysis and found that several oncogenic pathways, such as the oxidative phosphorylation, mTOR and MAPK signaling pathways, were dysregulated. The human DLD1 CRC cell line was obtained from the American Type Culture Collection (ATCC, Rockville, MD, USA) and cultured according to standard guidelines. All cells were authenticated by short tandem repeat fingerprinting before use at the Medicine Laboratory of the Forensic Medicine Department of Sun Yat-sen University (Guangzhou, China). All plasmids with the Flag-tag were provided by Saisofi Biotechnology Co., Ltd. (Jiangsu, China) and transfected into the cells using ViaFect Transfection Reagent (E4982, Promega, Madison, WI, USA) according to the recommended protocol. The lentiviruses packaging wild-type PTEN and site-directed mutants were synthesized by OBiO Technology (Shanghai, China). CRC cells were transfected with lentiviruses and selected with puromycin (HY-B1743A, MedChemExpress, NJ, USA) for 7 days.
