Bulk RNAseq analysis of in vitro differentiated wild type and Il24 -/- Th17 cells

Naïve T cells (CD3e+, CD4+, CD44-, CD62L+, CD25-) were isolated from splenocytes and lymph nodes of Il24 –/– mice (B6N(Cg)-Il24tm1.1(KOMP)Vlcg/2J) and Il24 +/+ wild type littermates (C57BL/6) using fluorescence-activated cell sorting, followed by three days of culture under Th17 skewing conditions (2 ng/ml TGF-ß, 50 ng/ml IL-6, with 2 µg/ml soluble aCD28 and 4 µg/ml plate-bound aCD3).
