Eosinophils costimulation with anti-Siglec-F and IL-33 synergistically increases their IL-4 and IL-13 release (Willebrand et al. PLoS One 2016). Here we analyze costimulation induced transcriptional changes involved that potentially enhance effector release. To determine transcriptional changes in primary eosinophils induced by anti-Siglec-F (clone E50-2440) and IL-33 costimulation we performed single and combined treatments with anti-Siglec-F and IL-33. Sorted eosinophils (SSChigh4get+) of IL-5tg_4get/C57BL/6 mice (original IL-5tg: Lee et al. 1997 J Immunol; original 4get: Mohrs et al. Immunity 2001) in which eosinophilia is driven by IL-5 and IL-4 producing cells are labelled with GFP were analyzed. We compare isotype, anti-Siglec-F, IL-33 and Siglec-F + IL-33 costimulated groups.
