Six to eight weeks’ old heterozygous Cas9 knockin animals (RosaCas9+/-) were administrated via tail vein injection of adeno-associated virus 8 (AAV8) expressing cre-recombinase and sgRNA targeting Mrg15. A firefly luciferase expression cassette was also included in the AAV vector to assist in vivo evaluation of delivery efficiency. AAV vectors with cre-recombinase and luciferase cassettes and no sgRNA were used as control viruses. Tissues were collected from animals under normal chow diet (NCD) after virus injection for 14 weeks for RNA-seq analysis.
